Responsibilities

  • Assist with vaccine development initiatives, including the production of vaccines for Phase 1 and 2 vaccine trials.
  • Independently develop scalable fermentation processes, and support purification processes for production of recombinant malaria antigens produced by Saccharomyces cerevisiae, Escherichia coli or Pichia pastoris and Myceliophthora thermophila.
  • Provide support to the Federal Task Leader (FTL) including but not limited to: small-scale and pilot-scale (60L) fermentation development.
  • Provide analysis of approaches for harvesting, purification development of scalable procedures.
  • Provide support to the analytical effort providing laboratory reagents for HPLC analysis.
  • Provide technical transfer of fermentation processes to a CDMO to support cGMP manufacturing and serving as the main Point-of-Contact.
  • Provide support in the development of design-of-experiment models for fermentation optimization and using Eppendorf fermenters and software, Raven methanol sensors, Akta Explorers, YSI Bioanalytical instruments and equivalent equipment to analyze products.
  • Perform various scientific experiments using Western blotting procedures, endotoxin testing and other HPLC/UPLC methods.
  • Record and analyze data using JMP software, Microsoft Word, PowerPoint, Excel and graphics programs.
  • Works with FTL to organize scientific data into a format that is suitable for manuscript publications to share discoveries with the scientific community.
  • Prepare various media, buffers, stock supplies, and other reagents for use in the laboratory, and maintain lab upkeep.

Qualifications

  • Minimum of a Bachelor's degree in a science related field with at least four (4) years of experience using Eppendorf Scientific fermentations systems at a variety of scales is required.
  • Minimum of four (4) years of related experience working in a scientific laboratory studying protein expression and purification is required.
  • Understanding of malaria and malaria vaccinology is highly desired.
  • Previous working experience at NIH is highly desired.
  • Experience at scale up, scale down fermentation processes using a variety of microorganisms especially Pichia pastoris and Myceliophthora thermophila C1.
  • Experience at the generation of ferritin nanoparticles using the novel expression system Myceliophthora thermophila C1 to produce novel drug products that can be used in preclinical and clinical evaluation studies.
  • Experience with Design of Experiments to optimize process parameters to identify optimal processing parameters to optimize cellular production of proteins used to create vaccines.
  • Excellent communication skills both in writing and speaking to share information with the FTL and other lab staff.
  • Familiar with using SharePoint, TEAMS, and Microsoft Office Suite programs.
  • Able to work alone and professionally as part of a team to accomplish assigned tasks in a timely manner.
