Discovery Research Vector Optimization Intern
Spark TherapeuticsAbout the role
Join the Spark Team
We were born of innovation, springing from the curiosity, imagination and dedication of remarkable scientists and healthcare visionaries. Our shared mission is to unlock the power of gene therapy to accelerate healthcare transformation.
We don’t follow footsteps. We create the path.
Our Research and Technology organization is currently seeking a Summer 2024 intern to join our Discovery team.
The intern will lead a project evaluating the interplay between bacterial genetics, plasmid sequence properties, critical quality attributes of plasmid DNA, and AAV manufacturing. Specifically, the student will test the applicability of inducible gene expression systems to control the activity of Cas9 gene editing modules and/or epigenetic modifiers, such as DNA methyltransferases, and the potential to use these enzymatic systems to control the properties of plasmids used in AAV manufacturing. The project will employ a variety of different experimental methods, including classic microbiology workflows, plasmid amplification methods, molecular biology methods (restriction digests, PCR), and next generation sequencing (NGS). Plasmids that have been successfully modified will be incorporated into AAV production platform to interrogate the impact of the modified sequences on AAV replication and packaging. This project will likely involve extensive cross-team collaboration at Spark, both within Discovery (bioconjugation team), and with Technical Development (Upstream PD).
- Must be a current student enrolled in an undergraduate or graduate program at an accredited college or university
- Minimum availability of 10 weeks (June 3 – August 9th)

- Participation in student organizations and/or other leadership experience
Preferred Qualifications
- Self-motivated and proactive in learning
- Demonstration of agile mindset
- Excellent verbal and written communication skills
- Willingness to lead
- Demonstrated interest in biotechnology and gene therapy industry
- Familiarity with microbiological and/or pipetting techniques
