About the role

Who You Are

In this role, the Scientist will be responsible for experiments to inform AAV construct design and selection in our Process Science and Research Core department. The scientist is responsible for planning and executing experiments to optimize production of plasmid DNA and AAV Vectors for research applications. The Scientist will have a working knowledge of early development of AAV gene therapy vectors from construct design through candidate selection. Job level is commensurate with experience.

 

How You Contribute

  • Plan and execute experiments to optimize molecular components of DNA and rAAV vectors
  • Design and lead research projects to improve the production of research grade plasmid and vector 
  • Design, execute and interpret early-stage rAAV vector screening experiments for construct optimization/selection and development of molecular tools for rAAV production
  • Design, execute and interpret experiments to better understand how AAV biology contributes to rAAV production 
  • Collaborate with Research, Process Development, and Analytical Development teams to implement experimental design changes and support internal research and program vector needs
  • Support lab operations, including inventory management, solution preparation, and standard lab tasks
  • Follow, propose improvements, and troubleshoot current laboratory protocols and operations 
  • Maintain accurate records and thoroughly document work through data entry into electronic laboratory notebooks, with minimal supervision
  • Prepare, organize, and track sample inventory during production and storage
  • Perform additional lab tasks as assigned

 

What You Bring to the Team

Education & Experience

  • PhD preferably in Science, Biotechnology, Engineering or other related field and 2+ years of hands-on lab experience in an industry setting OR
  • Master’s degree preferably in Science, Biotechnology, Engineering or other related field and 5+ years of hands-on lab experience in an industry setting
  • 2+ years of experience in design and evaluation of plasmids and molecular tools required for rAAV production

Competencies

  • Expertise in  gene therapy construct design and optimization is highly preferred
  • Hands on experience with mammalian cell culture and upstream rAAV production process is required
  • Hands on experience with research grade plasmid production and QC is required
  • Ability to learn and perform methods utilized in downstream purification of rAAV including ultracentrifugation and chromatography
  • Knowledge and hands-on experience utilizing proper lab technique while working with biologics (ex. bacterial or mammalian cultures, proteins, plasmid DNA, viral vectors)
  • Highly self-motivated, organized, and detail-oriented, with proven ability to foster strong relationships with other team members and stakeholders

  • Work is normally performed:   in laboratory environment  
  • Lifting requirements:   Under 20 lbs
  • Travel requirements:   None
  • Must have flexible work hours, and be willing to work outside of normal, scheduled hours; as necessary
